The Canadian swimmer qualified for the final in the women's 100-metre butterfly on Saturday by tying for fourth in the semifinals.
The top eight made the final.
Mac Neil is trying to become the first swimmer in history to win back-to-back Olympic titles in the event.
The London, Ont., native won silver in the 100-m butterfly at last year's world championships.
Gretchen Walsh of the United States set an Olympic record time of 55.38 seconds with compatriot Torri Huske finishing second at 56.00.
China's Yufei Zhang was third at 56.15.
Mac Neil and Germany's Angelina Koehler both finished at 56.55.
